What Is MOTS-c?
MOTS-c (Mitochondrial Open Reading Frame of the 12S rRNA-c) is a mitochondrial-derived peptide (MDP) – a signalling molecule encoded within the mitochondrial genome rather than the nuclear genome. Discovered in 2015 by Dr. Changhan David Lee at the University of Southern California, MOTS-c has rapidly become one of the most exciting peptides in the longevity and metabolic optimisation space.
Often called the “exercise mimetic peptide,” MOTS-c activates the same AMPK metabolic pathway that exercise triggers. It promotes glucose uptake, fatty acid oxidation, and mitochondrial biogenesis – essentially mimicking some of the cellular benefits of physical activity. This doesn’t mean it replaces exercise, but rather that it amplifies the metabolic benefits of your existing activity level.
How MOTS-c Optimises Metabolism
- AMPK activation: MOTS-c activates AMP-activated protein kinase (AMPK), the master metabolic switch that exercise triggers. AMPK increases glucose uptake into cells, promotes fatty acid oxidation, and stimulates mitochondrial biogenesis – the creation of new mitochondria.
- Mitochondrial function: As a mitochondrial-derived peptide, MOTS-c has a direct role in maintaining mitochondrial health and efficiency. It supports the electron transport chain and helps prevent mitochondrial dysfunction – a hallmark of aging.
- Insulin sensitivity: MOTS-c improves how your cells respond to insulin, enhancing glucose uptake and utilisation. This is particularly valuable for individuals with insulin resistance or metabolic syndrome.
- Exercise performance: By improving mitochondrial function and metabolic flexibility (the ability to switch between burning carbohydrates and fats efficiently), MOTS-c enhances both aerobic and anaerobic exercise capacity.
- Stress resistance: MOTS-c has been shown to translocate to the nucleus during metabolic stress, activating protective genes that enhance cellular resilience – a unique protective mechanism among peptides.
The Age-Related Decline of MOTS-c
Like many protective molecules, MOTS-c levels decline significantly with age. Circulating MOTS-c decreases as mitochondrial function deteriorates – creating a feedback loop where declining MOTS-c leads to worsening mitochondrial health, which further reduces MOTS-c production. Supplementation aims to break this cycle, restoring youthful mitochondrial signalling and metabolic function.
Research has shown that individuals with naturally higher MOTS-c levels tend to have better metabolic health markers, improved exercise capacity, and greater resilience to age-related metabolic diseases.
Who Benefits from MOTS-c?
MOTS-c appeals to athletes seeking performance optimisation, individuals over 35 noticing declining energy and metabolic flexibility, anyone with insulin resistance or early metabolic syndrome markers, and longevity-focused individuals who want to support mitochondrial health as they age. It pairs exceptionally well with a regular exercise programme.

How to Use Your Peptides
A complete visual guide to preparing, reconstituting, injecting, and storing your peptides safely. Follow each step in order.
Preparation
Gather your supplies: peptide vial, bacteriostatic water, syringes, alcohol swabs, cotton ball, and bandage.
Remove the plastic caps from both vials to expose the rubber stoppers.
Wipe the top of each vial with an alcohol swab. Let air-dry.
Reconstitution
Draw the required amount of bacteriostatic water into the syringe. Use our calculator if unsure.
Inject the water slowly into the peptide vial, letting it run down the inside wall. Do not spray directly onto the powder.
Gently swirl the vial until the powder dissolves. Do not shake.
Check the solution: no undissolved particles, no bubbles, clear liquid. If not, wait and swirl again.
Draw your dose slowly from the reconstituted peptide vial.
Injection
Hold the syringe needle-up, flick to move air bubbles to the top, and push the plunger until a small drop appears.
Choose your injection site: belly fat around the navel or outer thigh. Rotate sites between injections.
Clean the injection site with an alcohol swab.
Pinch a fold of skin at the injection site to lift the subcutaneous fatty tissue away from the muscle.
Insert the needle at a 45° angle into the subcutaneous fatty tissue, usually approximately half the needle length. Do not inject into muscle. Inject slowly.
Aftercare, Storage & Disposal
Withdraw the needle and press a cotton ball gently on the site for a few seconds.
Apply a small bandage.
Dispose of the needle in a sharps container.
Store reconstituted peptides in the fridge (2-8 °C). Avoid freezing and direct sunlight. Use within 90 days.
Category-Specific Dosing Guides
Typically administered once weekly. Because of how it’s studied, it is often discussed alongside regular physical activity.
Reconstitute
Add 2ml of BAC water to the 10mg vial (5mg/ml). Gently swirl the vial until the powder has fully dissolved.
Dosing
5–10mg once weekly via subcutaneous injection. Individuals researching or looking to buy MOTS-c online often administer their dose on a training day, either before exercise or shortly afterwards. This approach is based on its interaction with exercise-related metabolic pathways.
Cycle
Run for 4–8 weeks, then take 4 weeks off. Some users run 2–3 cycles per year. Regular resistance and cardiovascular training are commonly discussed alongside this peptide due to their shared relationship with metabolic and mitochondrial pathways.
